【摘要】:Isoasparticacidresiduesaregeneratedduringincubationofproteinsunderawidevarietyofconditionsinaqueoussolution.Suchresiduesaregeneratedmostcommonlythroughthedeamidationofaspargineresiduesalthoughsomereportsofisoasparticacidformationthroughtherearrangementofasparticacidresidueshavebeenpublished.Thepresenceofsuchresiduescanindicatethattheproteincontainingsuchresidueshassuffereddamagethatmayaffectthebiologicalactivityoftheprotein.Theprecisecorrelationbetweenthelevelofisoasparticacidcontentandthebiologicalactivityoftheproteinneedstobedeterminedonacasebycasebasis.Thetestmeasuresthelevelofisoasparticacidcontentinaproteinsample.Thislevelwilloftenbecorrelatedwiththedegreetowhichtheproteinhassuffereddeamidationatasparagineresidues.Inaddition,isoasparticacidresiduescanariseonoccasionthroughtherearrangementofasparticacidresidues.Forthesereasons,thelevelofisoasparticacidresiduesinproteinscanbeusedasageneralindicationthattheproteinsamplehassufferedsomelevelofdamageandshouldnotbeinterpretedtoindicatethepreciselevelofdamagetoanyoneregionwithinaproteinwithoutfurthertesting.1.1Thistestmethodcoversthedeterminationofisoasparticacidresiduesinaproteinorpeptidesample.Thistestmethodisapplicableforthedeterminationofisoasparticacidresiduesinasampleintherangeof2.5-50956;mol/L.Higherconcentrationscanbedeterminedfollowingdilution.Thereportedlowerrangeisbasedonsingle-operatorprecision.1.2ThevaluesstatedinSIunitsaretoberegardedasthestandard.1.3Thisstandarddoesnotpurporttoaddressallofthesafetyconcerns,ifany,associatedwithitsuse.Itistheresponsibilityoftheuserofthisstandardtoestablishappropriatesafetyandhealthpracticesanddeterminetheapplicabilityofregulatorylimitationspriortouse.
